摘要:
The aim of this study was to prepare an injectable sustained release hydrogel through drug polymer coupling model and Diels Alder click reaction
Puerarin hyaluronic acid coupling compound (PUE-HA) was prepared by connecting puerarin and hyaluronic acid through amide bond. PUE-HA and 4-arm maleimide polyethylene glycol (4-arm PEG-MAL) were crosslinked through Diels Alder click reaction to prepare injectable sustained-release hydrogel (PUE-HA-PEG) Compared with 1Furan/1MI and 1Furan/2MI hydrogels, the gelation time of 1Furan/4MI hydrogels can be shortened to 1 min. With the increase of MI ratio, the hydrogels have lower degradation rate (21 day degradation rate is less than 60%), better mechanical properties, and more obvious drug release effect
The experimental results show that the ratio of Furan/MI has a significant impact on the physical and chemical properties of injectable sustained-release hydrogels
本研究旨在通過藥物-聚合物偶聯物模型和Diels-Alder點擊反應制備一種可注射緩釋水凝膠.
將葛根素與透明質酸通過酰胺鍵連接得到葛根素-透明質酸偶聯物(PUE-HA),將PUE-HA和四臂馬來酰亞胺聚乙二醇(4-arm-PEG-MAL)通過Diels-Alder點擊反應交聯制備了可注射緩釋水凝膠(PUE-HA-PEG),并考察了三種不同呋喃(Furan)與馬來酰亞胺基(MI)比例對所得水凝膠的膠凝時間,形貌,溶脹度,降解度,葛根素體外釋放,水凝膠力學性能以及水凝膠生物相容性等理化性質的影響.與1Furan/1MI和1Furan/2MI水凝膠相比,1Furan/4MI的水凝膠膠凝時間可縮短至1 min.隨著MI比例的增加,水凝膠具有更低的降解速率(21 d降解率低于60%),更好的力學性能,緩釋釋藥作用更加明顯.
實驗結果表明,Furan/MI的比例對可注射緩釋水凝膠的理化性質具有重大影響.
