文獻:聚乙二醇-聚乙烯亞胺負載超順磁納米Fe3O4的合成及其基因轉染應用

A water-soluble graft copolymer polyethylene glycol grafted branched polyethylene imine (mPEG-g-PEI) was obtained by modifying polyethylene imine (PEI) with monomethyl ether polyethylene glycol (mPEG-OH), and polyethylene glycol polyethylene imine loaded with superparamagnetic iron oxide (PEG-PEI-SPIO) was prepared by ion exchange method. Then, α - carboxyl - ω - maleimide polyethylene glycol (mal-PEG-COOH) and multifunctional SPIO loaded antibody polyethylene glycol polyethylene imine (Ab PEG PEI-SPIO) were synthesized. The composite of PEG-PEI-SPIO and plasmid DNA (pDNA) had a particle size of about 105 nm. In vitro experiments showed that T cells could specifically absorb... The Ab-PEG-PEI loaded SPIO targeting group showed good performance in gene transfection experiments, and real-time non-invasive observation can be performed using magnetic resonance imaging during the gene transfection process
采用單甲基醚聚乙二醇(mPEG-OH)改性聚乙烯亞胺(PEI),得到水溶性接枝共聚物聚乙二醇接枝支化聚乙烯亞胺(mPEG-g-PEI),并利用離子交換法制備了負載超順磁性氧化鐵的聚乙二醇-聚乙烯亞胺(PEG-PEI-SPIO).然后合成了α-羧基-ω-馬來酰亞胺聚乙二醇(mal-PEG-COOH)和多功能負載SPIO的抗體-聚乙二醇-聚乙烯亞胺(Ab-PEG-PEI-SPIO).PEG-PEI-SPIO與質粒DNA(pDNA)的復合物(Ab-PEG-PEI-SPIO/pDNA)粒徑約105 nm.體外實驗結果顯示:T細胞能特異性地吸收Ab-PEG-PEI負載的SPIO,靶向組在基因轉染實驗中有較好的效果,在基因轉染的過程中可用磁共振手段進行實時無創觀測.
